To establish a convenient assay for p53 mutation detection, we investigated the concordance of p53 status determined by immunohistochemical staining and the real-time RT-PCR amplification of mRNA of TP53 gene with seven different primers covering all known mutations of p53 gene in ovarian cancer. A total of 98 archival paraffin-embedded tissues from ovarian cancer patients were examined and their p53 status was also confirmed by immunohistochemical staining. Using a real-time RT-PCR protocol with three different primer sets, we identified the presence or absence of p53 mRNA of TP53 gene. The concordance rate of p53 mRNA expression with p53 immunohistochemistry was excellent with three primer sets: Co1, Co5 and Co7 (99% each, P = 0.005, Bonferroni method) although the primers for Co1 were the most sensitive and specific for detecting p53 mutations. The overall sensitivity of this assay for p53 mutation was almost 100% with this combination of primers, and the proportion of patients with p53 mRNA in different grades of differentiation (G) by immunohistochemistry (IHC) was significantly different (P = 0.03). In addition, there was no significant difference in the frequency of p53 mutations between the groups of borderline and invasive ovarian cancer in terms of different stages and grades.
